Here’s my take to start things off.
Let’s begin with the candidates who’s popularity is skewed by outside factors.
Cuphead and Mugman: They’re the faces and the protagonists of the franchise, it’s impossible for them to not leave a strong impression on the players’ memories.
Inkwell Isle 1 Bosses: Statistically speaking, the earlier parts of games are played more than the later parts. This means early bosses are more strongly remembered than later ones.
Next, we have what I consider the ‘honorable mentions’.
Grim Matchstick: The green dragon with a stutter appears to be the most frequently represented boss from Inkwell Isle 2. Whether it’s because of his design, his personality, or his gameplay is up to debate.
Cala Maria: I don’t really need to say anything more here, do I?
And now it’s time for the two characters I think people like the most, divided into the ‘Expected’ and ‘Unexpected’ categories.
Expected: King Dice. This guy is a triumph of character design. His appearance, personality, behavior, animation and theme music all fit together perfectly. The moment you enter the first Die House, you realize “this character is going to be a fan favorite, no question.”
Unexpected: Chalice. Somehow a ghost who only ever appeared three times in order to give you a new powerup became a DLC playable character and part of the main cast in the cartoon adaptation. That sort of thing doesn’t just happen without a reason.
Also, I wanted to mention my King Dice headcanon:
Unlike everyone else, King Dice’s work for the Devil is completely voluntary. He became ‘the Gamest in the Land’ entirely through his own ability and determination, and had everything he could ever want, temptation wise. He walked into Hell and made the Devil a deal: he’d use his skills and charisma to aid Satan in stealing people’s souls, and in return, King Dice would get… to use his skills and charisma to aid Satan in stealing people’s souls. King Dice is so evil and cruel that he sees being able to damn people to Hell as it’s own reward.
This, to me, is something that would diminish his character if it wasn’t there. He, out of everyone fought in the game, truly has no excuse for doing bad things. There was nothing compelling him to become evil, but he did anyway, just because he could.
Personally my favorite is king dice, (of course) but my second favorite is phear lap! I feel as if that horse boy doesn't get enough attention.

Dr. Kahl- the brothers bump into him when they lose a lucky coin in the junkyard. He agrees to build a metal detector to help them find it, but then decides the device isn’t good enough and goes crazy trying to add “perfections” to it. Eventually he builds the giant robot and the brothers have to stop him before things get further out of hand. Also the robot should be voiced by Vin Diesel, the original voice of the Iron Giant.
Beppi the Clown- the brothers revisit the Carnevil, only to find that it has been abandoned by the Devil and refurbished by Beppi, a bubbly and very friendly clown. However, the new carnival just isn’t quite as fun, but the brothers don’t have the heart to tell this happy clown that his carnival sucks. Beppi notices their disdain and struggles to keep them entertained, eventually getting furious and breaking his happy character. It turns out the Devil hired him to lure Cuphead back to get his soul, and Beppi doesn’t want to fail. A chase scene ensues, featuring the Carnival Kerfuffle track.
Mangosteen- the brothers are touring Beppi’s carnival and come across a series of “wondrous” creatures that are clearly faked (“baby pegasus” is a pony with cardboard wings, “worlds largest rat” is obviously a capybara, etc.). Beppi then introduces Mangosteen as “the real magic 8 ball” and Mangosteen flashes their signature grin. The brothers wave them off as a lame attraction, and Mangosteen frowns. It’s only a short cameo, buts it’s a reference to the setup the Devil used when trying to get Cuphead to take off the sweater.

My favorite boss design is probably King dice,
He is very cool looking and I definitely prefer his design more in the game than his softer appearance in the show. However! He was my favorite character in the show I thought his song was great and he was really really funny!
